RANKL/RANK/OPG Triad
Osteoblasts produce RANKL (receptor activator of NF-κB ligand), which binds RANK on osteoclast precursors, driving osteoclastogenesis and bone resorption. Osteoblasts also produce OPG (osteoprotegerin), a soluble decoy receptor that binds and neutralizes RANKL. The RANKL/OPG ratio determines net resorption tone. Denosumab is a clinical anti-RANKL antibody.
Inflammation Drives RANKL
TNF-α, IL-6, and IL-1β all upregulate RANKL while suppressing OPG. Postmenopausal osteoporosis (estrogen loss → cytokine elevation → RANKL up) and rheumatoid arthritis (synovial cytokine overload → erosion) both involve RANKL excess. Phycocyanin's anti-inflammatory effects reduce RANKL transcription by 25-40% and increase OPG by 15-25% in chronic inflammation models.
Conclusion
Spirulina shifts RANKL/OPG ratio toward bone preservation through inflammation suppression. Clinical relevance: postmenopausal bone loss prevention, RA-associated bone erosion reduction, and metabolic bone disease support.
